In this project, we aim at elucidating fundamental gene regulatory networks during hematopoietic stem cell (HSC) differentiation at the single-cell level. We have recently established an inducible culture system that is capable of examining the molecular networks during B cell lineage commitment from HSCs (Ikawa et al., 2015). Additionally, the digital RNA-seq analysis has also been developed using barcoding strategy (Shiroguchi et al., 2012). By combining these systems, we will dissect the dynamic expression patterns of key regulators including transcription factors, lncRNAs and epigenetic modifiers during HSC fate determination at the single-cell level.
