Single Molecule measurement of Nanog during the transition from ESC to EpiSC

Pluripotency of mouse embryonic stem cells (mESCs) is maintained by gene regulatory network including core transcription factors such as Nanog, Oct4, and Sox2. While the molecular basis has been revealed along with development of Omic-technologies, the physical aspects of those has not been clarified yet. A few papers had reported the single molecular behaviour of transcription factor. The molecular species however were limited because of the practical problem of the single molecular measurements. We here try to measure the diffusion, the binding kinetics, and the nuclear transports at single molecular level in single cells.
