Technology development for single-cell RNA sequencing with a novel RNA amplification method. We will improve the two critical points in whole transcript amplification for single-cell transcriptome. 1) Sensitivity for detection of transcripts and 2) easy-to-use with microfluidics devices. This method provided a 10-100 fold increase in cDNA yield over that obtained with commercial kits, making it possible to perform gene expression profiling for low-expression genes with single-cell RNA, thus expanding the number of detectable genes. We will also implement single-cell RNA-seq with the novel RNA amplification method on a commercially available microfluidics device. Our team will widely share our method to solve various biological questions in the project.
