The need for Immune-cell repertoire profiling (i.e., characterization of variable regions of T-cell receptor and/or B-cell receptor) is rapidly growing, especially in the clinical research field to identify repertoires associated with human diseases. After many requests from medical institutes in Japan, we decided to launch a platform in RIKEN IMS to provide Ig/TCR repertoire analysis pipeline. In this pipeline, we includes single-cell culturing system, immune-cell repertoire sequencing with Next-generation DNA sequencer, .high-throughput system for expression plasmid construction, and characterization of recombinant BCR/TCR (in collaboration with RIKEN QBiC. The constructed pipeline will be open to the research community in Japan and our final goal is to identify key human repertoires valuable for therapeutic intervention and/or diagnosis in two different human diseases and vaccine development.
